摘要
目的 观察STAT3在小鼠胚胎神经管发生和神经上皮分化迁移阶段的时空表达规律,为探讨STAT3在胚胎神经系统发生和发育过程的作用及可能机制提供形态依据。方法 采用全胚胎原位杂交技术和免疫组织化学技术,观察STAT3在神经胚形成和神经上皮分化迁移阶段的表达。结果 ①E8.75d整个鼠胚几乎没有观察到STAT3mRNA的表达,在E9.5d ,STAT3mRNA在前脑泡急剧增多,到E10 .5d ,阳性信号出现在中脑泡,E11.5d时在后脑泡的一狭窄区域有STAT3mRNA的表达;②E13 .5d ,在侧脑室周围即未来大脑新皮层区域、间脑、发育中的视网膜、第四脑室周围的脑组织(未来的脑桥)、脊髓、三叉神经节和背根神经节C1均可观察到较强的STAT3蛋白阳性信号,E14 .5d和E15 .5d时,大脑皮层、间脑、视网膜、第四脑室周围以及发育中的脑桥和脊髓等部位观察到中等强度的STAT3蛋白表达。结论 STAT3可能参与了神经管的关闭和神经上皮的分化和迁移。
Objective To observe the expression of STAT3 in the process of neurulation, the differentiation and migration of the progenitor cell of mouse. Methods The expression of STAT3 was observed by whole-mount in situ hybridization and immunohistochemistry. Results ①At E8.75 d, STAT3 mRNA was hardly detected in the embryo; at E9.5 d, STAT3 mRNA increased in the prosencephalon dramatically; at E10.5 d, STAT3 mRNA was observed in the mesencephalon; at E11.5 d, STAT3 mRNA was limited to a narrow region in the metencephalon. ②At E13.5 d, the positive signal of STAT3 protein expression was detected in the surrounding region of the lateral ventricle developing retina, diencephalons, pons, the surrounding region of the fourth ventricle, medulla oblongata, trigeminal ganglion, dorsal root ganglion of C1 etc. At E14.5 d and E15.5 d, it was found that the expression of STAT3 protein decreased slightly in the pallium, diencephalons, developing retina, the surrounding region of the fourth ventricle, pons, and spinal cord etc. Conclusion STAT3 might be involved in the closure of the neural tube and the differentiation and migration of the progenitor cell.
