摘要
目的研究人心房肌细胞内钙离子(Ca2+)浓度与心房颤动(AF)的关系。方法急性分离风湿性心脏病(RHD)伴或不伴有AF患者以及先天性心脏病(CHD)窦性心律患者的心房肌细胞,用Fluo3作为钙指示剂,用激光共聚焦显微镜技术测定细胞内Ca2+浓度。结果在RHD患者中,伴有AF者的心房肌细胞内Ca2+浓度(517±98nmol/L)明显高于不伴有AF者(262±65nmol/L,P<0.01),而二者均明显高于CHD患者的心房肌细胞内Ca2+浓度(117±48nmol/L,P均<0.01)。结论RHD患者心房肌细胞内Ca2+浓度升高。RHD伴AF患者心房肌细胞内存在Ca2+超载。
Objective To study the relationship between intracellular calcium concentration of atrial myocytes and atrial fibrillation (AF) in humans. Methods Atrial myocytes were isolated from right atrial appendages of rheumatic heart disease (RHD) patients with or without AF, and also isolated from that of congenital heart disease (CHD) patients with sinus rhythm. Intracellular Ca 2+ concentration was measured with the fluoresent Ca 2+ indicator Fluo-3 and laser scanning confocal microscopy. Results Intracellular Ca 2+ concentration of RHD patients with AF was significantly higher than that of non-AF RHD patients [(517±98) nmol/L vs (262±65) nmol/L, P<0.01]. Both of them were signifiCantly higher than that of CHD patients with sinus rhythm [(117±48) nmol/L] (all P<0.01). Conclusion Intracellular Ca 2+ concentration was increased in RHD patients. Ca 2+ overload was found in atrial myocytes of RHD patients with AF.
