青岛地区人疱疹病毒6型毒株的分离与初步鉴定 被引量:1

摘要 ①目的 分离培养人疱疹病毒6型(HHV -6)青岛地方株并进行初步鉴定。②方法 取20例青岛地区健康献血员唾液标本,接种至预激活的脐带血单个核细胞(CBMC)中进行病毒分离培养。出现典型细胞病变(CPE)的标本采用电镜观察,并提取病毒DNA,应用聚合酶链反应(PCR)扩增 HHV -6 基因组保守区特异性片段,然后应用限制性核酸内切酶技术进一步鉴定。③结果 唾液分离培养得到1例具有典型CPE的标本,电镜观察显示细胞内存在具有疱疹病毒特征的病毒颗粒,酶切产物电泳结果显示66 bp和97 bp两条带,初步鉴定为HHV -6B毒株。④结论 成功分离培养得到1例HHV- 6青岛地方株,为进一步研究青岛地区 HHV- 6 的流行及致病性奠定了实验基础。 Objective To isolate and identify human herpesvirus 6(HHV-6) local strains of Qingdao. Methods The saliva specimens of 20 healthy blood donors in Qingdao were inoculated in cord blood mononuclear cells(CBMC). The isolates with typical cytopathic effect(CPE) were preliminarily identified by electron microscope, polymerase chain reaction(PCR) and restricted endonuclease. Results The typical CPE appeared in one of 20 samples. Some herpesvirus-like virions could be observed in the cells with typical CPE by electron microscope. The HHV-6 specific DNA fragment was amplified by PCR and two DNA fragments, 66 bp and 97 bp, were gained by restricted endonuclease digestion, suggesting that the isolates was HHV-6B strain. Conclusion The isolation of local strain of HHV-6 in Qingdao will be helpful in further study of epidemiology and pathogenicity of HHV-6.
