摘要
目的 :分离和鉴定与小鼠合子基因组激活 (ZGA)以及母源调控向胚胎调控 (MZT)过渡事件相关的基因片段 ,为进一步认识ZGA和MZT发生的分子机制奠定基础。方法 :利用单个植入前胚胎抑制性消减杂交 (SPE SSH)的方法 ,对单个小鼠MⅡ卵母细胞和 2 细胞期胚胎进行双向消减杂交 ;利用cDNAarray对获得的基因片段作进一步的鉴定。结果 :从构建的消减杂交库中随机挑取得 31个克隆 ,经cDNAarray鉴定发现其中有 5个MⅡ期特异表达的基因片段和 9个 2 细胞期特异表达的基因片段 ,Genbank检索发现其中 1 0个片段是首次报道在植入前发育阶段表达。结论 :这些基因片段具有明显的期特异性表达的特点 ,可能在ZGA、MZT和胚胎植入前发育过程中起着重要的作用。
Isolation and identification of gene fragments related to zygote genome activation(ZGA) and maternal zygotic transition(MZT) occur in mouse preimplantation embryos at 2 cell stage. Method: single preimplantation embryonic suppression subtractive hybridization(SPE SSH) was used to isolate stage specific expressed sequence tages(ESTs) of mouse oocytes and 2 cell embryos and cDNA array was used to identify these differential expressed ESTs. Result: After oocyte stage and 2 cell stage enriched SSH libraries constructed,5/31 fragments were proven to be specifically expressed in MⅡ oocytes and 9/31 in 2 cell embryos by cDNA array method.To our knowledge,10/14 stage specific expressed fragments were firstly reported to involve in preimplantation development.Conclusion: cDNA array of whole preimplantation stage further proven that these genes might play an important role in ZGA,MZT,and preimplantation developemnt.
