摘要
目的:探讨人外周血淋巴细胞遭受H2O2损伤时,8-羟基-2'-脱氧鸟苷三磷酸酶(8鄄oxo鄄dGTPase)的表达及端粒长度的变化,评估氧化应激对端粒长度的影响。方法:体外培养人外周血淋巴细胞,分为正常对照组、氧化组和抗氧化组,采用半定量逆转录聚合酶链反应(RT鄄PCR)及凝胶成像技术,测定8鄄oxo鄄dGTPase的mRNA表达水平,并应用Southern杂交技术检测端粒长度的变化。结果:①随着H2O2作用时间的延长,8鄄oxo鄄dGTPasemRNA的相对含量也增加,尤以氧化组增加最显著(P<0.05),而抗氧化组与对照组相比较则差异无显著性(P>0.05);②端粒长度则随着时间的变化表现为缩短,且3组之间均有差异(P<0.05)。结论:①8鄄oxo鄄dGTPase的表达随着氧化应激的程度加深而显著增加;②维生素C具有一定的抗氧化作用,它能明显减低过氧化损伤,抑制8鄄oxo鄄dGTPase的生成,同时能减缓端粒缩短的速率;③氧化应激在一定程度上可以引起人淋巴细胞端粒长度的缩短。
Objective:To observe the variation of 8-oxo-dGTPase expression and telomere length of human peripheral blood lymphocytes caused by oxidative stress and to evaluate the effect of oxidative stress in telomere length. Methods:Human lymphocytes were cultured in vitro and divided into three groups:oxidative, anti-oxidative using Vit. C and normal control group. RT-PCR were applied to study the mRNA expression of 8-oxo-dGTPase and southern hybridizing were applied to determine telomere length. Results:①The expression of 8-oxo-dGTPase was increased significantly in lymphocytes damaged by H2O2 and there was no significant difference between anti-oxidative and normal control group. ②Telomere length was shorten with the change of time. All the three groups had significant difference(P < 0.01). Conclusion:①The expression of 8-oxo-dGTPase was increased significantly with increasing levels of DNA damage, and pre-treatment of cells with Vit.C can inhibit the expression of 8-oxo-dGTPase induced by oxidative stress; ②Pre-treatment of lymphocytes with Vit.C can slow the rate of telomere shortening; ③Oxidative stress takes some reaction on telomere shortening.
