氯沙坦对TGF-β_1在自发性高血压大鼠肾小管表达的影响及作用 被引量:3

摘要 目的 :研究转化生长因子 β1(transforminggrowthfactor β1,TGF β1)在自发性高血压大鼠 (sponta neouslyhypertertensiverat,SHR)肾小管的表达及其与肾小管间质纤维化的关系 ,氯沙坦对TGF β1在肾小管表达的影响及肾保护作用。方法 :以同龄雄性正常大鼠 (WistarKyoto ,WKY)和SHR为研究对象 ,其中SHR大鼠分为阳性对照组和氯沙坦组。监测治疗前后大鼠尾动脉血压、肾功能及 β2 微球蛋白(β2 MG) ,并采用免疫组织化学的方法检测TGF β1在肾小管中的表达。结果 :同WKY组比较 ,β2 MG在阳性对照组显著增高 (P <0 .0 5 ) ,氯沙坦组较阳性对照组显著减少 (P <0 .0 1) ,而尿素氮和血肌酐各组间差异无显著性 (P >0 .0 5 )。TGF β1在WKY组肾小管的表达无或极微量 ,在阳性对照组肾小管的表达显著 ,且随高血压病程的进展 ,TGF β1的表达显著增加 (P <0 .0 5 ) ,而TGF β1在氯沙坦组的表达显著减少 (P <0 .0 1) ,血压亦显著降低。结论 :TGF β1在自发性高血压大鼠肾小管的表达显著增加 ,它可能是致肾间质纤维化的重要因素。氯沙坦具有降低血压、减少蛋白尿、保护肾功能及延缓肾小管间质纤维化的作用。 AIM: To study the expression of transforming growth factor β 1(TGF β 1) in the nephric tubule of spontaneous hypertensive rats and the relation of the TGF β 1 and the renal tubulointerstitial fibrosis, and to investigate the effect of losartan on this expression and the protection effects of it on the kidney. METHODS: Same age male WKYs and SHRs were involved in this experiment. SHRs were divided into positive control (SHR C) group and losartan treatment (SHR L) group. The artenia pressure, renal function and β 2 MG of mice was monitored before and after experiment and the expression of TGF β 1 in nephric tubule were measured by immuonohistochemitry assay. RESULTS: Compared with the WKY group, the contents of β 2 MG markedly increased in SHR C group (P< 0.05 ) and decreased in SHR L group (P< 0.01 ), but the contrast of BUN and Scr not significance changed in both two groups (P> 0.01 ). No or a few expression of TGF β 1 occurred in WKY group. The expression of TGF β 1 significantly increased in the positive control group with the progression of the hypertensive course, and meantime, the expression of TGF β 1 significantly decreased in the losartan treatment group and the blood pressure also decreased. CONCLUSION: As the expression of TGF β 1 intensively increased in hypertensive course, TGF β 1 may be the important factor leaded to the renal tubulointerstitial fibrosis. Losartan has the effects on lowering blood pressure, reducing albuminuria, protecting renal function and delaying the renal tubulointerstitial fibrosis.
