摘要
以混合溶剂法直接用对硝基苯甲醛和吡咯合成了Meso 5 ,10 ,15 ,2 0 四 (对硝基苯基 )卟啉 (H2 TNPP) ,通过SnCl2 /浓HCl还原得到Meso 5 ,10 ,15 ,2 0 四 (对氨基苯基 )卟啉 (H2 TAPP) .D 生物素经羰基二咪唑活化后 ,在吡啶中与H2 TAPP反应合成生物素 卟啉偶联物 ,偶联物与醋酸锌通过固相反应 ,制得了金属配合物 ,并通过FT IR ,UV Vis ,MS ,1HNMR和元素分析对生物素 卟啉偶联物的结构进行表征 .合成的偶联物可望为生物医学检测提供一种稳定的生物探针 ,用于肿瘤的早期诊断或其他生物检测 .
Meso 5, 10, 15, 20 tetra (p nitrophenyl) porphyrin (H 2TNPP) was synthesized by pyrrol and p nitrobenzaldehyde in the mixed solvents composed of propionic acid, acetic acid and nitro benzene. A reduction to Meso 5, 10, 15, 20 tetra (p aminophenyl) porphyrin (H 2TAPP) in a SnCl 2/concentrated HCl system was conducted. Using Carbonyldiimidazode as condensing reagent, D biotin can be activated to couple with H 2TAPP in pyridine to from biotin H 2TAPP; In order to get a metalloporphyrin complex, biotin H 2TAPP was treated with Zn(Ac) 2 by a solid state synthesis. The structures of the above synthetic products have been characterized by FT IR, UV Vis, MS, 1HNMR and elementary analysis. The biotin H 2TAPP metallocomplex is hoped to provide a stable biologic probe for biomedicine, in aid of the diagnosis of the early tumour and other biological detection.
