PPAR-γ2基因P12A变异与2型糖尿病的相关性研究 被引量:4

摘要 目的: 研究过氧化物酶体增殖物激活受体-γ2 (PPAR-γ2 )基因外显子B的P12A变异与2 型糖尿病(T2DM)及其相关临床特征的关系。方法: 选取新疆地区维吾尔族154 例,其中糖耐量正常(NGT)组91 例,T2DM组63例,应用聚合酶链限制性内切酶片段长度多态性方法对2 组的基因型进行测定;对2 组的临床变量,包括身高、体重、腰围、臀围、空腹血糖(FBG) 、血清胰岛素(FINS)及血脂进行测定;对PPAR γ2 基因P12A变异的基因型与各临床变量的关系进行研究。结果:T2DM组和对照组PPAR γ2 基因P等位基因和A等位基因的分布频率分别为87. 3%、89. 6%和12. 7%、10. 4%, PP 基因型频率为74. 6%、79. 1%, PA+ AA 基因型频率为25.4%、20.9%,2组基因型及等位基因频率差异均无统计学意义(P>0.05)。T2DM组A12 变异携带者与腰围有关(P=0.019)。本组维吾尔族人PPAR γ2 基因P12A变异的A等位基因分布频率与白种人群相近,与中国汉族人差异较大。结论:维吾尔族PPAR γ2基因外显子B的P12A变异与T2DM无明显关系,但A12携带者可能影响或加重T2DM患者的脂代谢异常。 Objective: To investigate the association study of P12A variant in peroxisome proliferators activated receptor-γ_2 (PPAR-γ_2) gene with type 2 diabetes and its relative clinical characteristics. Methods: The genotypes of P12A variant in PPAR-γ_2 were determined by polymetase chain reaction restriction fragment length polymorphisms assay in 154 subjects of the Uygur population in Xinjiang of China, including 91 subjects with normal glucose tolerances and 63 type 2 diabetes mellitus patients; the clinical characteristics including height, weight, waist, hip, FBG and FIN of the subjects were detected and the realationship between the genotype of P12A variant and the clinical characteristics were analysed. Results : The allele frequencies in the case and control group were 87.3%, 89.6% for P and 12.7%,10.4% for A. the genotype frequencies were 74.6%, 79.1% for PP, 25.4%, 20.9% for PA+AA. The P12A variant of PPAR γ_2 was not significantly associated with type 2 diabetes (P>0.05) . The frequencies of the Ala12 carrier of in diabetes patients was associated with increased waist circumference(P=0.019). The frequencies of PPAR-γ_2 gene in Uygur population were similar to the caucasian and different from the Han population. Conclusion : The data show that P12A variant polymorphism in Uygur population is not significantly associated with type 2 diabetes, but it may affect or enhance the metabolic disorder of the type 2 diabetes patients.
