摘要
介绍一种简便、快速的血清过氧化氢酶活性的比色测定方法。探讨了其比色波长的选择、H2O2、钼酸铵试剂浓度与吸光度的关系,溶血、脂血、黄疸标本对测定结果的影响,以及标本与试剂的存放等因素。本法精密度试验批内变异系数(cv)为4.6%,批间CV为5.0%。对47例新生儿脐血清标本的过氧化氢酶活性测定平均值是:147.7±59.9KU/L(42.0~236.4).测定了977例健康人的血清过氧化氢酶活性为:52.7±17.0KU/L(15.2~118.2)。试验结果表明:男性血清过氧化氢酶活性较女性16.6%,其结果有随年龄增长而增高的趋势。
A rapid,sensitive spectrophotometric
assayfor determination of serum catalase activity is described.The correlations of the
absorbance with the factors includ-ing choice of colometric wavelength, the concen tra tion
ofhydrogen peroxide and ammonium molybdate, and the stor-age of samples and reagents is
studied, Hemolytic ,lipemicand icteric sera increased the absorbance without influmoeon the
catJase assay. Due to the high content of catalasein erythrocytes,artificial hemolysis of the
sample would in-crease the catalase activity of serum.The method has an in-tra- and interassay
precision(CV)of 4.6%and 5.0%, re-spectively. In 47 healthy neonatus the niean and SD val-ues
of the catalase activity of umibilical cord serum were147.7±59.9KU/L(42.0~236.4).IN 977
healthy indi-viduals the mean and SD Values of serum catalase activitywere 52.7±17.0
KU/L(15.2~118.2) with 16.6%higher in males than in females. The serum catalase activi-ty
increases wit age.
