摘要 目的探讨抗乙酰胆碱受体单克隆抗体A7(抗-AChR mAb A7)对实验性自身免疫性重症肌无力(EAMG)的作用. 方法经Lewis大鼠腹腔注射5 mL 20倍浓缩的含抗-AChR mAb A7培养上清液,建立EAMG被动转输模型.对照组经腹腔注射同体积的PBS.每8 h监测体重和临床症状评分.48 h后处死实验动物.采用放射免疫法检测AChR,并计算出AChR损失率.结果抗-AChR mAb A7诱导的EAMG模型AChR损失率高达24.3%~45.2%,而且AChR损失率与临床症状评分相关(r=0.74, P<0.01).抗-AChR mAb A7攻击位点在终板的AChR上.结论抗-AChR mAb A7可用于诱导EAMG模型及有关研究. Objective To investigate the effect of anti-acetylcholine receptor monoclonal antibody A7 in experimental autoimmune myasthenia gravis (EAMG). Methods Passive transfer EAMG was induced by i.p. injection of 5 mL 20 fold concentrated culture supernatant of A7 in lewis rats. Control rats were injected i.p. with 5 mL PBS. Monitored body weights and assessed clinical signs were scored every eight hours. Animals were killed 48 h after injection of the mAb A7 and the loss of acetylcholine receptor (AChR) was demonstrated by measurement of the muscle AChR concentration with radioimmunoassay (RIA) and calculated the AChR loss rate. Results There were higher AChR loss rate found in EAMG induced by mAb A7, from 24.3% to 45.2%, which were correlated to clinical signs score (r=0.74, P<0.01). The target site of mAb A7 was in AChR of end-plate. Conclusions Anti-AchR mAb A7 could be used to induce EAMG model for investigating the pathogenesis of ~myasthenia gravis.
