摘要
目的:研究丁酰化壳聚糖丝裂霉素药膜在兔眼房水中的释放性能。方法:应用高效液相色谱法分别在0.25,0.5,1,2,3,6,24h行前房穿刺术,抽取房水测定植入兔眼巩膜瓣下的丁酰化壳聚糖丝裂霉素药膜中MMC在房水中的含量。设对照组,为另1眼巩膜表面放置MMC棉片(MMC0.25g/L)5min,NS冲洗。结果:植入药膜组0.25,0.5,1,2h后的房水中可检测出MMC,含量22.5~130.3μg/L(最低检测浓度10μg/L)对照组仅在0.25,0.5h的房水中检测到MMC。结论:丁酰化壳聚糖丝裂霉素药膜植入兔眼小梁切除术的巩膜瓣下具有一定的缓释作用。
AIM: To explore the release of butyrylchitosan MMC film in rabbit aqueous humor.· METHODS: A direct injection method for determination of MMC in ocular aqueous of different rabbit groups after trabeculectomy by restricted-access media (RAM)-HPLC was developed. Butyrylchitosan MMC film was used in the right eyes of the rabbits and tampon soaked with 0.25g/L MMC was used in their left eyes as controls for 5min and washed by Saline. Different concentrations of MMC in rabbit ocular aqueous were determined at 0.25, 0.5, 1, 2, 3, 6 and 24h after trabeculectomy respectively.· RESULTS: In Experimental Group, the MMC concentration of butyrylchitosan MMC film could be determined at 0.25, 0.5, 1 and 2h after trabeculectomy, and the concentration ranged from 22.5 to 130.3μg/L. In the control group, It could be determined only at 0.25 and 0.5h after trabeculectomy. · CONCLUSION: Butyrylchitosan MMC film can decrease the release and decomposition of MMC in trabeculectomy.·
