摘要
原生质体是研究植物遗传工程的好材料。自从Cockingt(1960)首次用酶解法从蕃茄根尖分离出大量原生质体以来，高等植物原生质体的研究已经开展得比较广泛，但由于技术上的原因，目前从原生质体诱导出完整植株的还只限于烟草、矮牵牛、石刁柏，油菜和马铃薯等十余种。而藻类方面的工作则报道甚少。
The present paper describes some of the experiments on the preparation, culture and fusion of somatit cells from U.lnza and M.angicava.The main results are as follows.(1) protoplasts of the two species were obtained by means of treating the thalli with hypertonic enzyme solution ,shaking them by a sudden decrease osmotic pressure and washing them with hypertonic seawater medium. The optimnm conditions of preparing the protoplasts of both species were investigated.(2) After a 20-day's culture in the MPES medium added with KT, IAA,Fe++ and Vit.C, each of most Protoplasts underwent cell division ard developed into autotrophic cell mass.Tubular germlings were induced by treating the masses with the MPES medium diluted with distilled water for 24 hours and then culturing them in the MPES medium added with Fe ++.Normal U.linza grew up under a bacteria-free condition.It was also observed that the ; rotoplasts isolated from the sporophytes of U, linza could only regenerate into sporophytes and those from gametophytes only into gametophytes.(3) The effects of different concemtrations of KT,IAA,Fe++ and Vit. C on promotirg cell division ard indusing plant regeneration were examined ny the orthongoal experimental method. The freque cies of success is green algae were ob iosly higher than in any higher lants.(4) The fusion of the protop asts of U.linza and M.angicava which was labe'ed with 0,01% neutral red was induced by PEG. The intergene-ric hybrid cells were isolated and cultured which formed into autotrophic cell masses.
