摘要
将猪瘟兔化弱毒疫苗阳性血清与猪瘟野毒株HeBZJK1.95、GZGD1.95、HeBHH2.95、BJXC1.95、HeNBY1.96、BJCY1.96、BJSY2.96、BGTX3.96、HeNXH2.98、FJFQ1.99、JL1.94和标准石门株在PK-15单层细胞上进行中和试验,测定了猪瘟病毒抗体在体外对野毒株的中和作用能力.结果表明,阳性血清对HeBZJK1.95、GZGD1.95、B.JSY2.96、JL1.94和石门株的中和效价为1:256,对BGTX3.96株为1:128,对HeBHH2.95、BJCY1.96、HeNXH2.98株为1:64,对BJXC1.95株为1:16,对FJFQ1.99株为1:8,对HeNBY1.96株为1:4.由此表明,对不同毒株血清中和效价不同.另外测定了猪体内免疫抗体与免疫保护力的关系.采用1头份猪瘟兔化弱毒疫苗主动免疫没有母源抗体的猪,免疫后14 d和192 d抗体滴度<1:16,攻击野毒HeNXH2.98株,仔猪能够完全保护.采食初乳的2组仔猪,母源抗体滴度为<1:16~1:64,分别免疫2头份和4头份猪瘟兔化弱毒疫苗,免疫后抗体滴度均下降1~2个滴度(1头母源抗体滴度低的仔猪,免疫4头份后由<1:16达到1:16).攻击野毒HeNXH2.98株后,免疫2头份的仔猪,不管免疫后抗体水平多高(<1:16~1:64),均不完全保护;免疫4头份的仔猪,免疫后抗体滴度较低(1:8)时,不完全保护;抗体滴度≥1:16时,能够完全保护.动物免疫保护试验表明,1头份疫苗免疫抗体水平能起到良好攻毒保护作用,但母源抗体会影响疫苗免疫抗体水平和攻毒保护效果.
The neutralization tests of HCLV serum with hog cholera wild virus HeBZJK1.95,GZGD1.95,HeBHH2.95,BJXC1.95,HeNBY1.96,BJCY1.96,BJSY2.96,BGTX3.96,HeNXH2.98,FJFQ1.99,JL1.94,HCVSM.97 strains had been done on PK15 cells.The results indicated that neutralization antibody titers to HeBZJK1.95,GZGD1.95,GZGD1.95,BJSY2.96,JL1.94,HCVSM.97 strains were 1∶256,to HeBHH2.95,BJCY1.96,HeNXH2.98 strains were 1∶64,to BJXC1.95 strain was 1∶16,to FJFQ1.99 strain was 1∶8,to HeNBY1.96 strain was 1∶4.The neutralization antibody titers were not identical to different strains.The relativity between immune antibody and animal immune protection had been detected.The pigs without getting colostrum had been vaccinated 1 dose of vaccine were able to protection against challenge with HeNXH2.98 strain,although antibody titers <1∶16.The antidody titers of two group pigs getting colostrum dropped 1-2 titers after had been vaccinated 2 doses and 4 doses of vaccine.The pigs after had been vaccinated 2 doses of vaccine and that the antibody titer was 1∶8 after being vaccinated 4 doses of vaccine were not able to protect against challenge with HeNXH2.98 strain.The pigs that antibody titers were ≥1∶16 after being vaccinated 4 doses of vaccine were able to protect against challenge with HeNXH2.98 strain.The results of animal immune protection indicated that the antibody level after being vaccinated 1 dose of vaccine could lead to effective protection against challenge with HeNXH2.98 strain,but was affected by maternal antibody.
