紫草籽抗氧化物质(SOD和黄酮类化合物)的研究 被引量:3

摘要 紫草籽SOD提取工艺为 :粗酶液提取 ,超滤 ,硫酸铵分级沉淀 ,DEAE -FF离子交换层析和SephadexG - 10 0凝胶过滤。实验表明 ,纯化后的SOD比活达到 32 89.2U mg .pro ,活性回收为 35 .4 %。紫草籽中黄酮类化合物的最佳提取条件为 :乙醇浓度6 0 %、固液比 1:5、温度 5 0℃、提取时间为 2h。然后用HZ816树脂进一步分离纯化得精制黄酮 ,其纯度为 86 .3% ,总黄酮回收率为 77%。进一步实验表明精制黄酮对DPPH·的EC50 为 12 .6mg L ,优于茶多酚 ,与维生素C非常接近。紫草籽精制黄酮对小鼠离体肝、肾和心组织中的MDA生成均有抑制作用 ,其中对离体肝组织中MDA生成的抑制效果最明显。 The SOD from Zicao seeds was separated and purified by the methods of extracting the crude enzyme, ultra filtration concentration, (NH 4) 2SO 4 fractional precipitation, ion-exchange chromatography (DEAE-FF) and gel-filtration. The result showed that the specific activity of SOD was improved to 3289.2U/mg.protein, and its yield was about 35.4%. The optimum extracting conditions of flavonoids from Zicao seeds were decided as follows: 60% ethanol, solid/liquid ratio 1:5, extracting for 2h under temperature 50℃. The crude flavonoids could be further purified by the HZ816 macroporous resin and the content of total flavonoids was 86.3%, with a total flavonoids recovery rate of 77% .The EC 50 of the flavonoids on DPPH· was 12.6 mg/L, lower than tea polyphenol and close to ascorbic acid.The flavonoids had obvious effect on inhibiting tissue homogenate MDA. By comparison of the inhibitory rate of tissue homogenate MDA of heart, liver and kidney, it was found that its effect on liver was the best .
