血清IgG、IgA、IgM、C_3和C_4四种测定方法的方法学评价 被引量:3

METHODOLOGIC EVALUATION OF FOUR METHODS FOR MEASURING IgG、IgA、IgM、C_3 and C_4 IN SERUM

摘要 本文介绍血清IgG、IgA、IgM、C_3和C_4四种测定方法的方法学评价试验。结果表明:简易免疫比浊法,微量免疫比浊法和超微量免疫比浊法测定前述五项指标的准确度,精密度和灵敏度均高,且都比单向放射免疫扩散法好。并把原简易免疫比浊法中的反应时间从37℃60分钟改为37℃30分钟,其吸光度仍稳定;把原简易免疫比浊法中的测定波长从495nm改为340nm,原超微量免疫比浊法中的波长从492nm改为450nm,这样可以提高灵敏度。我们用这四种方法同时测定了60份献血员血清IgG、IgA、IgM、C_3和c_4水平,并对这四种方法做了干扰试验。由于免疫比浊法操作简单、快速,且准确度、精密度和灵敏度均好,因此,其不失为测定血清IgG、IgA、IgM、C_3和C_4较好的方法。 Four methods for measuring IgG,IgA,IgM,C3 and C4 in sera were evaluated.The results showed that the accuracy,precision and sensitivity of simple,micro-and ultramicro-immune turbidimetry for measuring the five items all are high and better than that of the single radial immunodiffusion method. The reaction temperature and time of the simple immune turbidimetry was changed from 37℃60 minutes to 37℃ 30 minutes,showing stable absorbance;for improving their sensitivities the wavelengths of the simple immune turbidinictryand the ultramicro-immune turbidimetry were changed from 495nm to 340nm and from 492nm to 450nm respectively.using these four methods, IgG, IgA,IgM,C3and C4 of 60 donors' sera were determined and interference tests also were done on all of them,The immune turbidimetry is simple,rapid ,accurate,precise and sensitive,and it is of choice for measuring Serum Igs,C3ang C4.
