SPA-ELISA评价三种不同纯度牛源细粒棘球蚴囊液抗原在包虫病免疫诊断中的价值

摘要 本文采用SPA-ELISA法评价了三种不同纯度牛源细粒棘球蚴囊液抗原在包虫病免疫诊断中的价值,用三种抗原检测了41例包虫病患者血清中特异IgG抗体的水平,它们的阳性率分别为:牛源细粒棘球蚴液粗抗原86.0%,半饱和硫酸铵法纯化抗原91.4%,氯化镁—磷钨酸法纯化抗原92.3%,两种纯化抗原与粗抗原的阳性检出率相比,经统计学处理具有显著差异,认为西北地区牦牛体内寄生的细粒棘球蚴囊液抗原不乏抗原活性;氯化镁—磷钨酸法和半饱和硫酸铵法是两种简便易行,纯化效果理想的抗原纯化方法,宜于在基层推广应用。 By using three different biochemical methods, we obtained three antigens of different purity from bovine hydatid cyst fluid. SPA-ELISA tests were performed by using these three preparations (1. B. EgcF. Ag, 2. B. EgcF. Ag (NH4) 2SO4 and 3. B. EgcF. AgMgCl2 ) as antigens to determine the presence of corresponding antibody (IgG) in the sera of operation-proved cases, as well as those from normal adults. Our study shows that the positive percentages of B. EgcF.Ag, B .EgcF .Ag MgCl-2 and B.EgcF. Ag(NH4) 2SO4 are 86%,92.3% and 91.4%,respectively. In the light of SPA-ELISA results, the reactions of the three different antigens to 41 cases of human cystic hydatidosis proved statistically significant. It was also found that the cross-reactions between the purified antigens and crude antigens decreased. In conclusion, we suggest that: 1. ) The antigenicity of EgcF.Ag from yaks herded in North-West district is the same as that of other hosts.2.) The two methods are worthy to popularize in lots of primary medical units.
