摘要
以自制高活性PPK为材料,在氰基硼氢化钠存在下经水溶性乙醛酸修饰其表面氨基,使其抗不可逆热失活稳定性有显著提高。结果表明,修饰PPK在等电点等基本性质方面都有变化。修饰PPK的BAEE活性为天然酶的82%,氨基修饰度为58%,抗蛋白酶水解,贮藏和冻干稳定性都有加强。
Reductive alkylation of surface amino groups in high activity PPK (pig pancreas kallikrein) by glyoxylic acid in the presence of sodium cyanoborohydride results in increasing the stability against irreversible thermoinactivation. Analyses show the essential properties of modified PPK were changed slightly. The medified PPK comparing with natural PPK is decreased 18% in the BAEE activity; Modified degree of the surface amino groups is 58%. The stability in lyophilization and storage and the ability in antiproteinase hydrolysis of modified PPK were increased.
