摘要
将表皮生长因子及神经生长因子分别作用于分离所得的鼠胚成纤维细胞及其转化细胞的细胞核。结果表明表皮生长因子可以提高游离细胞核中DNA拓扑异构酶的活性,但神经生长因子无此作用。
Epidermal growth factor (EGF) and nerve growth factor can stimulate DNA synthesis in quiescent cultured NC-3H10 (normal C_3H10T_(1/2) C18) and transformed counterpart TC_3H10 cells, Now the mechenism is unkown. In this paper, according to the method described by Grenfell ,the nuclei were purified from NC_3H10 and TC_3H10 cells. Nuclei prepared by this method were virtually free of intact cell and cytoplasm. Nuclei were stimulated by EGF or NGF in the topoisomerase reaction buffer.It was found that EGF can increase the activity of topoisomerase ,but NGF can not. The stimulation of topoisomerase by EGF may represent a key step in the process by which EGF induces DNA synthesis and cell division.
