摘要
研究神经节苷脂GM3(简称GM3)对部分纯化的人红细胞膜Ca2+-ATP酶的作用和对完整兔红细胞浆[Ca2+]的影响。结果表明:外源性GM3对Ca2+-ATP酶的作用呈浓度依赖性双相调节即浓度为1~6.5μmol/L时抑制酶活性,浓度大于6.5μmol/L时激活酶活性;GM3不影响钙调素(CaM)对酶的激活;CM3对兔红细胞浆[Ca2+]也呈浓度依赖性双相调节即在GM3浓度低于69μmol/L时[Ca2+]有不同程度升高,浓度大于60μmol/L时则降低[Ca2+]。
The effects of exogenous gangliosides GM3 on partially purified Ca2+-ATPase of human RBC and on cytosolic calcium of intact rabbit RBC were studied.Results reveal that: ① Exogenous GM3 Showed a concentration-dependent,biphasic effect on Ca2+-ATPase activity. Low concentrations (1~6.5μmol/L) of GM3 could inhibit Ca2+-ATPase while high concentrations of GM3(higher than 6.5 μmol/L) could activate the enzyme.GM3 was not able to reduce the stimulating effect of calmodulin on the enzyme. ② Exogenous GM3 was also showed biphasic function on cytosolic calcium concentration [Ca2+] in RBCs.When the concentration of GM3 was lower than 60μmol/L, [Ca2+] was increased to various degrees, but was decreased when GM3 concentrations were higher than 60μmol/L.
