摘要
自淋球菌4.2Kb质粒pJDl基因序列选择一对引物序列合成引物,PCR扩增产物为230bp片段。经30次循环后,最低可检出10CFU。特异性试验结果表明,这对引物与白念珠菌有部分同源性,PCR扩增产物小于75bp。与脑膜炎奈瑟菌及其他13种常见的细菌、4种念珠菌、沙眼衣原体、解脲支原体、正常女性宫颈分泌物中的细菌均无同源性。临床标本检测结果表明,与涂片法和培养法相比较,阳性检出率可达100%。试验表明,应用本方法可直接快速检测分泌物中的淋球菌。
One primer set was selected from the published complete nucleotide sequence of 4.2 kb cryptic plasrmid pJDI of N.gonorrhoeae.The amplified product of PCR was 230bp fragment.which could be detected by agarose gel electrophoresis.The sensitivity of PCR was 10 CFU after it was performed for 30 cycles.The results of PCR assay showed the primer set exhibited partial homology to candida albicans with an amplified product less than 75bp and showed no homology to Neisseria meningititis and other 13 bacteria spp.,Candidia spp.,Chlamydia trachomatis,Mycoplasma urealyticum, and bacteria in cervical discharges of the normal women.The PCR results of the clinical specimens showed that the sensitidty was 100% comparing with Gram stain method and culture method.It is indicated that Neisseria gonorrhoeae in clinical specimens can be rapidly detected with PCR technique.
