摘要
在只经戊二醛未经锇酸固定的洋葱和百合小孢子母细胞核的超薄切片中,看到一种由颗粒和纤维组成的网络结构。减数第一次分裂前期的早期,这种结构分布于整个细胞核内的非染色质区和核仁腔隙,并与染色质和核仁相连,随着细胞的发育,分布在非染色质区的颗粒和纤维的密度逐渐变小,当细胞进入中期以后,这些颗粒和纤维结构消失而染色体之间的网络结构却依然清晰可见。该结构经胃蛋白酶的消化处理后消失,经ETDA倒退染色后不能被漂白。本研究结果提示,植物花粉母细胞核基质可能也包含有一定的纤维网络结构,该结构的功能可能与染色体的组装,核形态的维持有关。
In the nuclei of microsporocytes of lily (Lilium daviddi) and onion (Allium cepaL.) which have undergone the primary fixation of gluteraldehyde but no osmic acicd postfixation, a matrix network made of fibrillar and granular materials has been seen under the electron microscepe. This matrix network is located in the non-chromatin regions and nucleolar interstices, and associated with chromosomes, nucleoli and nuclear membrane in early prophase. With the development of the cells. the fibrillar and granular structures are getting thinner and thinner, and by Metaphase I it has been found by the ETDA regressive staining that they have transferred to the surface of chromosomes. The structures can be eliminateed by pepsin, but not bleached by the ETDA regressive staining. The present result suggeststhat the nuclear matrix structures similar to theones found in many kinds of amimal cells may also exist in the pollen mother cells of higher plants and its function is probably related to the maintenance of nuclear shape, the location of chromosomes and nucleoli,and the reorganization of chromeosomes.
