摘要
以菘蓝子叶和下胚轴为外植体进行组织培养,筛选出植株再生的适宜的培养基为MS+KT2mg/L+NAA0.2mg/L。对器官发生进行系统的细胞组织学观察发现:菘蓝下胚轴培养,细胞启动和不定芽的形成随下胚轴部位的不同而有所不同;不定芽发生的数量与其维管轴的发育有关,整个下胚轴分化不定芽的数量呈梯度变化。
Plantlet was obtained from cotyledon and hypocotyi of Isatis indigotica Fort.invitro, In the experiments, the highest frequency of bud induction occured in MS medium sup-plemented with 2 mg/L KT and 0.2m/L NAA, Segments from three distinct regions of thehypocotyl were used.Histocytological observations indicated that the number of bud forma-tion was related to the develpment of their fibrovascuear bundles and that the number of budformation from the top to the end of the hypocotyl showed gradient changes.In addition,theroot-tip chromosomal numbers of plantlets were examined。
