摘要
用安格斯牛7日龄冻胚16枚,在液氮中保存894d后,用36℃水浴解冻(25S),全部回收。在室温27℃条件下,用10%蔗糖一步脱除抗冻剂(甘油)10min后,移入20%犊牛血清PBS10min,再移人同样浓度的犊牛血清PBS中10min,按常规形态鉴定方法.对胚胎评级,10枚为可用胚胎,解冻后胚胎可用率为62.5%,将10枚可用胚胎用非手术法,移入发情周期与胚胎发育期同步的5头受体牛,结果2头妊娠,移植妊娠率为40%。妊娠足月后,2头受体牛各产公犊1头,试验结果表明,在液氮中保存894d之久的安格斯牛冻胚,进行洲际间引种能够成功,这也是安格斯牛冻胚洲际间引种,在我国首次获得成功。
Sixteen Angus seven-day frozen embryos were imported from Australia. After being Stored in liquid nitrogen for 894 days, these embryos were thawed in 36℃ water bath for twenty-five seconds and recovered totally. Under temperature 27℃, 10% sucrose solution was used to remove cryoprotectant for ten minutes, then embryos were transferred into 20% fetal calf serum PBS for ten minutes, and another ten minutes in the same PBS.With regular identification method, embryos were graded. Ten transferable embryos were. recovered and transferable percentage was 62. 5% after thawing. Ten good embryos were transferred non-surgically into five recipients synchrollous to embryonic age.As a result , two recipients were pregnant and the pregnancy rate was 40%. Each of the two recipients gave birth to one single male calf. The Experimental results indicated that Angus frozen embryos transcontinental import could succeed under preservation,in liquid nitrogen for 894 days. Also,this is the first successful transcontinental Angus embryos import in China.
