低温逆境中水稻幼苗质膜ATP酶和5′─核苷酸酶活性的变化 被引量:6

摘要 本文以氯化钾(CeCl_3)沉淀的细胞化学方法,观察了水稻幼苗细胞在低温胁迫中质膜ATP酶和5′-核苷酸酶活性的变化,得到的主要结果如下:(1)在适宜的生长温度(28℃)下,水稻幼叶细胞内的ATP酶主要定位于质膜,细胞间隙和胞间连丝等处,其中以质膜和细胞间隙处的酶活性最强;当幼苗经过1℃的低温处理后,质膜上的ATP酶活性比适温生长时明显降低。(2)生长在适宜温度(28℃)下的水稻幼苗,其细胞内的5-核苷酸酶活性专一性地定位于质膜和胞间连丝上,表现出很强的活性反应;幼苗在1℃低温处理2天后,其细胞内的5-核苷酸酶活性仍是严格定位于质膜及胞间连丝上,只是酶活性比低温处理前明显降低。这些结果进一步证明,质膜是低温伤害的最初部位。 In this paper,the activity changes of ATPase and 5-nucleotidase in the cells of the rice seedlings under low temperature stress were studied using cytochemical method of cerium trichloride precipitates。The main results are summarized as follows:(1) When rice seedlings grew at the optimum growth temperature(28℃), the ATPase in the cells of the rice seedlings was mainly localized in plasmolemma,plasmodesmus,and intercellular spaces,but only those AT-Pases, which situated in plasmolemma and intercellular spaces,mariifested high activities。When rice seedlings were treated at the temperature of 1 ℃ for 48 hrs,the ATPase activities in plasmolemma decreased considerably compared with those of the rice seedling growing at the optimum temperature。(2)When rice seedlings grew at the optimum temperature,the 5-nucleotidase in the cells of the rice seedlings uniquely located in plasmolemma and plasmodesmus,and showed high activities。The uniformly distributed deposits formed a black and clearly identifiable ring round the protoplasts;This indicates that the 5-nu-cleotidase,being used as an indicator of plasmolemma,is fit for not only animal materials,but plant materials also。When rice seedlings were treated at the temperature of 1℃ for 48 hrs,though the 5′-nucleotidase were still firmly localized in plasmolemma and plasmodesmi,its activities decreased significatly compared with those of non-treated rice seedlings,there only sparsely appeared some black deposits in plasmolemma for the treated seedlings。These results further indicate that plasmolemma is the primary site of low temperature stress。
