摘要
分析经γ射线离体照射的正常人外周血单个核细胞(PBMC)及急性放射病恢复期患者PBMC中T细胞受体(TCR)、CD_3、CD_(18)及CD_(25)阳性细胞。发现经1~6Gyγ射线照射后立即分析,正常人上述4种标志阳性T细胞百分率无明显改变;受照射细胞经体外诱导培养4~14d,TCR、CD_3及CD_(25)阳性细胞百分率不同程度呈照射剂量依赖性下降,随培养时间延长增加,CD_(18)阳性细胞百分率的变化幅度相对较小。免疫细胞化学分析显示,照射后8~12d,T细胞胞浆内有较多TCR(β)及CD_3阳性物质出现。提示:辐射致TCR、CD_3阳性T细胞减少可能与细胞内TCR及CD_3表达和装配异常有关。
Using a flow cytometry(FCM),we detected and enumerated the positive cell percentage of T cell receptor(TCRα/β),CD_3,CD_(18)and CD_(25) in normal human peripheral blood mononuclear cells(PBMC)irradiated by γ ray in vitro and PBMC from persons who suffered acute radiation symptoms 3 years earlier.The results showed that after PBMC from the normal persons were irradiated with 1~6 Gy dose the positive cell percentage of TCR,CD_3,CD_(18)and CD_(25) did not change significantly when analysis was conducted immediately after PBMC were irradiated,while the positive cell percentage of TCR,CD_3 and CD_(25) decreased with the increase in irradiation dose and increased with prolongation of culture time significantly at different levels.The change in positive cell percentage of CD_(18) was relatively smaller than that of the others after irradiated PBMC were cultured in virto for 8~14d. Immunocytochemical analysis showed that more TCR and CD_3 positive stains presented in cytoplasm 8~12d after irradiation.These results implied that the decrement of positive TCR and CD_3 T cells induced by irradiation might be related to abnormal expression and disabled assembly of TCR/CD_3 complex.
