胸腺素增强白细胞介素Ⅱ介导的活化淋巴细胞抗瘤活性研究

摘要 本文作者研究了胸腺素(Thymosin,TH)对重组白细胞介素Ⅱ(rIL_2)诱生活化杀伤细胞(Lymphokine Activated Killer cells,LAK)的影响。试验采用正常人及肝癌患者的外周血淋巴细胞(Peripheral Blood Lymphocyte,PBL)为效应前体细胞,以新鲜人体肝癌细胞为靶细胞,并与K_(562)红白血病细胞株做了比较。结果显示:经低剂量rIL_2(300U/ml)与TH联合活化的效应细胞,其杀肿瘤细胞活性明显高于等剂量rIL_2单用组(方差分析P<0.05),而与大剂量rIL_2(1000U/ml)单用组效应相近;胸腺素与rIL_2联合应用的增效强度不受植物血凝素(phasin,PHA)的影响,且在肝癌患者和正常人PBL之间及以新鲜人体肝癌细胞或K_(562)细胞株为靶细胞之间的差异无显著性意义。这提示TH可增强rIL_2介导的LAK活性,且此增效作用具有广谱性和可重复性。 The authors had investigated the effects of thymosin (TH) on the rIL_2-inducedLAK activity in vitro by using the exclusion assay of trypan blue. Peripheral bloodlymphocytes (PBL) from healthy volunteers And patients with live cancer were used.Fresh human liver cancer cells and erythroleukemic cell line K_(562) were used as tumortarget cells. The results indicated that the level of lysis caused by lower dose of rIL_2plus TH was significantly higher than that by the same dose of rIL_2 alone, and wasthe same as that by higher dose of rIL_2, alone. The enhancing effect of TH on rIL_2-induced LAK activity were obtained both in the presence or absence of phytohemag-glutinin. As between fresh human liver cancer cells and K_(562) cell line, and betweenhealthy volunteer's PBL and patients' PBL, no significant diffenences were display-ed. These results indicated that TH is able to enhance the rIL_2-induced LAK acti-vity repeatedly and reliably.
