摘要
以[^3H]地塞米松为配体,应用Scatchard分析测定30例糖尿病患者白细胞糖皮质激素爱体的最大结合容量及平稀解离常数。结果表明1型和Ⅱ型患者白细胞GR的RO均明显降低,且亲和力也降低植增加,就用Boyden小室法检测白细胞趋势化移动及氢化可的松对白细胞ChtM的抑制率。结果表明白细胞GR改变的同时们有效率指标F对白细胞ChtM的FI的相应变化,说明糖尿病患者白细胞GR的降低具有临床意义。
The maximal binding capacity (Ro) and dissociation constant (Kd) of glucocorticoid receptors (GR) in peripheral leukocytes were estimated in 30 diabetic patients (type Ⅰ 10 cases.type Ⅱ 20 cases) by redioligand binding assay. Ro and Kd of GR in diabetics and controls were 3 827+658 sites/cell and 5 274±1 033 sites/cell (P<0.01)and 7. 95±2. 63 nmol/L and 6.14±2. 64 nmol/L(P <0. 05)respectively. It was demonstrated that Ro was significantly lower and Kd was higher in the diabetics than in the controls. The leukocytic chemotactic migration (Cht M)was determined by Boyden Chamber. Cht M in diabetics and controls were 86. 2±11. 4 μm and 101. 5±7. 8μm respectively (P<0. 01). These results indicated that fraction inhibition (FI) of Cht M by hydrocorti-sone was 21. 4±3. 6 μm and 24. 0±4. 4 μm respectively(P<0. 05). There was a positive correlation between the FI and GR. It is demonstrated that change of GR has clinical significance.
