摘要
本文用HPLC 法测定氟康唑的血药浓度,血样用乙酸乙酯提取,以ODS 为固定相,乙腈—0.05mol/L 磷酸二氢钾为流动相,非那西丁为内标,在260nm 波长定量检测,在0.25~10.00μg/ml 范围内线性关系良好,重现性好,回收率为101.4~101.6%(n=8)。用本方法测定了10名健康志愿者口服150mg 氟康唑胶囊的体内血药浓度,并进行了药物动力学参数计算。
Fluconazole is a novel triazole antifungal agent.A sensitive HPLC method has been developed for the determination offluconazole in serum.Phenacetin was used as the in ternal standard.Fluconazole was extracted from alkalized serum with ethylacetate and evaporated to dryness.HPLC analysis was carried out by using ODS column and acetonitrile-0.05mol/Lpotassium biphosphate as the mobile phase.The detection wavelength was 260nm.Calibration curve Was linear between 0.25and 10.0μg/ml.The assay procedure has been applied to monitoring serum levels in oral pharmacokinetic study in tenvolunteers.Parameters were calculated.
