摘要
应用PCR技术扩增人ZFX和ZFY基因特异性DNA顺序,在男性DNA中检测到ZFX基因特异和ZFY基因特异的两种扩增产物,在女性DNA中仅检测到ZFX基因特异的一种扩增产物。采用ZFX/ZFY扩增系统进行性别鉴定的优点为:①检测X、Y两条性染色体,无假阴性结果;②ZFY基因位于Y染色体短臂,不受Y染色体长臂正常变异影响;③ZFY基因邻接TDF基因,适于性反转受检者;④扩增产物可经电泳直接区分。
A PCR amplification system for simultaneous detection of the human ZFX and ZFY genes have been designed. In this system, one common upstream primer and two gent-specific downstream primers were used. Two distinguishable bands were detected in male DNA samples after gel electrophoresis. of the two PCR products represented by the two bands, one was specific for the ZFX gene and another was for the ZFY gene. And in female DNA samples, only one ZFX-specific band was observed. This system could be applied to sex determination of variable biological materials. The false-negative and false-positive results in the previously reported systems for sex determination could be overcome.
