摘要
应用脾直接免疫方法制备HCC单克隆抗体,获得了高效价BALB/c小鼠抗血清。稀释到1:150倍仍为强阳性(ELISA法)。采用上述方法用HCG免疫小鼠的脾细胞与NS-1细胞融合生成的杂交瘤细胞所分泌的抗体效价为1:6000以上(ELISA法)。脾直接免疫方法与传统免疫方法相比较,操作简便,免疫时间短,节省抗原。利用此方法获得分泌特异性抗体的杂交瘤与同一抗原,按传统方法免疫动物获得的杂交瘤所分泌的抗体,在效价上和抗体特异性上没有差别。是一种很实用的免疫方法。
Anti-HCG monoclonal antibodies were prepared by intrasplenic direct immunization. The titer of the antiserum detected by FLISA was above 1:150, and the titer of the monoclonal antibody detected by the same method was above 1:6000. This method were simple and convenient to handle, and the dose of antigen and time were both saved compared with common immunization method. It had no difference in titers and specifities of the McAbs obtained between direct and common immunization method.
