摘要 从SD 大鼠肾小管刷状缘提取了致肾炎性的肾小管上皮抗原(Tub-Ag)。用此抗原免疫白兔获得的抗血清注入大鼠,复制出被动型Heymann 肾炎(PHN)动物模型。此后,应用兔抗Tub-Ag 的抗体进行鼠肾动脉灌注研究,并用免疫电镜观察了大鼠肾中Tub-Ag 的存在部位。结果发现:灌注的抗体能与肾小球基底膜(GBM)上皮侧及裂隙膜结合。致肾炎性的Tub-Ag 主要定位在足突基都细胞膜、裂隙膜、近曲小管上皮细胞顶部凹褶及微绒毛基部。 In the experiment,the nephritogenic tubular epithelial antigen(Tub-Ag)from renal tubularbrush border of SD rats was isolated and the animal model of passive Heymann nephritis(PHN)was successfully reproduced in the rats after injection of the antiserum made by immunizing rab-bits with the Tub-Ag.Then,the purified antibody against Tub-Ag was perfused into rat renalartery and the uitrastructural localization of Tub-Ag in rat kidney was investigated with immu-noelectronmicroscopy.It was found that the perfused antibody could bind to the epithelial side ofGBM and the filtration slit membrane in absence of CIC.Moreover,the nephritogenic Tub-Agwas demonstrated to be mainly located in the cell membrane of the basal portions of the foot pro-cesses and in the filtration slits of podocytes,and it was also detected on the apical invaginationsand at the base of microvilli in proximal tubule epithelial cells.
