摘要
采用透明质酸特异结合蛋白(Hyaluronate binding protein,HABP)为透明质酸(Hyaluronate,HA)抗体,建立了血清HA 的夹心ELISA 检测方法。关键步骤为HABP 的提纯及其酯化生物素的标记,并利用亲合素生物素系统放大提高其灵敏度。临床初步应用提示,尿毒症和慢性肝病患者血清HA 含量明显高于正常人。本方法简便、快速、重复性好,灵敏度可达2.5ng/ml,可适用于HA 的临床和基础研究。
A hyaluronic acid binding protein(HABP)was isolated from bovine nasal cartilage,and a
immunoenzymoassay of hyaluronate (HA) has been developed.Plastic microtest plates were
coated with a 10mg/L solution of HABP and HA bound to HABP on the solid phase was reveled
by biotin-conjugated HABP.The assay was applied to determine HA in sera and the specificity
was demonstrated by hyaluronidase digestion.The intrassay variation coefficient was 6.2—
14.3%,interassay variation coefficient was 6.9—8.8%.In 40 blood donor sets,HA was found
at 11.15±5.4μg/L.Patients with uremia and chronic liver diseases showed a higher serum HA
level than that of healthy controls.The ELISA method may be useful to basic and clinical studies
of HA.
