摘要
2月龄的Wistar大鼠。经颈静脉注射^3H-Ru486(^3H-R)或^3H-孕酮(^H-P)后,可见自显曩银粒在子宫肌层细胞核内有明显的聚集,而腔上皮和腺上皮内银粉分布相对较少。在阴道、输卵管肌层细胞核内也有聚集。垂体前叶细胞的核、浆以及下丘脑、视上核、室旁核、弓状核等核团内神经元的胞核内都有相似的银粒聚集。所有动物的隔肌内均未见自显影银粒定位。用非标记Ru486,孕酮或地塞米松竞争的结果表明,Ru486在下丘脑、垂体、子宫等水平均和孕酮竞争特异性结合部位,很可能是孕酮受体结合部位。这提示Ru486在体情况下对上述各个水平的生理机能均可能产生影响,其抗早孕和诱导月经可能是通过多个环节而起作用的。
Twenty-one castrated oestrogen-primed Wistar rats were injected via the jugnlar vein with [ 3H ] Ru486 or [ 5H ] progesterone. The 4μm frozen sections of the samples were processed for thaw-mounted autoradiography and showod that the nuclear concemration of radioactivity Was most distinct m muscular and stromal cells of the uterus, and the epithelial nuclei of lumina and glands showed weak labelling. Nuclear localization was also observed in muscle cells of the vaqina, cervix, oviduct and in the nuclei and cytoplasm of anterior pituitary cells. In the hypothalamus, specific localization of radioactivity existed in neurones accumulated in the preopotic nucleus, preoptic suprachiasmatic nucleus and the periven-tricular nucleus. No localization was found in the diaphragm. pretreatment with Ru486, but not with dexamethasone, reduced the nuclear concentration of radioactivity of [ 3H ] progesterone in the vagina, uterus, oviduct, pituitary and hypothalamus. The nuclear concentration of radioactivity after injection of [ 3H ] Ru 486 was also decreased by prein-jection with progesterone. The autoradiographic results suggest that Ru 486 and progesterone competed for the specific binding site ( possibly a progesterone receptor ) in the target cells at the levels of the uterus, pituitary and hypothalamus in vivo.
