摘要
目的 :研制、评价 5 -Br-PADCAP直接光度法测定血清锌试剂盒。方法 :在表面活性剂存在下 ,用 2 - (5-溴 - 2 -吡啶偶氮 ) - 5 - [(N ,N -二羧基甲基 )氨基 ]苯酚 (5 -Br-PADCAP)作显色剂手工分析法和自动分析法测定血清锌。结果 :该法线性范围 0~ 80 μmol/L ,手工分析法和自动分析法平均回收率为 99 8%和 10 0 3% ,批内变异系数(CV)和批间变异系数分别为 0 0 2 1、 0 0 16和 0 0 2 9、 0 0 2 4 ,与原子吸收分光光度法比较具有良好的相关性 ,线性回归方程和相关系数分别为Y =1 0 0 4X - 0 2 84 ,r =0 995 2和Y =0 9917X - 0 175 ,r =0 996 8。 82例健康人血清锌含量分别为9 0 2~ 2 2 34μmol/L和 8 4 9~ 2 2 14 μmol/L ( x± 2s)。 结论 :用 5 -Br-PADCAP直接光度法试剂盒测定血清锌方法简便、灵敏可靠 ,适合临床应用。
Objective:To investigate and evaluate the reagent kits of analysis of the zinc in serum using 5-Br-PADCAP direct spectrophotometric method.Methods:For manual and automatic determination of serum zinc with 5-Br-PADCAP in the presence of surfactant.Results:The manual method and automatic method were all linear in the range of 0~80 μmol/L,the average rate of recovery were 99.8% and 100.3%,the withinrun and between run CV were 0.021、0.016 and 0.029、0.024.Comparing two methods(Y) with AAS(X),we obtained the regression equations:Y=1.004X-0.284,r=0.9952 and Y=0.9917X-0.175,r=0.9968.The reference interval(±2s)(n=82).Conclusions:This reagent kits of direct spectrophotometric analysis of the zinc in serum are simple,sensitive and suitable for clinical application.
