摘要
目的 探讨中药治疗自然流产中滋养细胞肝素表皮生长因子(HB-EGF)、核增殖抗原(PCNA)的表达及 相关性分析对滋养细胞增殖的影响。方法 中药治疗溴隐亭致SD大鼠流产模型组,另设黄体酮、外源性泌乳素 (PRL)治疗模型;单纯模型、正常孕鼠、非孕鼠为对照组,各12只;于孕12 d处死观察妊娠结局。通过RT-PCR 及免疫组化观察各组胎盘、子宫内膜HB-EGFmRNA及PCNA的表达,电镜观察微绒毛变化。结果 中药与黄 体酮、外源性PRL治疗组及正常孕鼠组胎盘HB-EGF、和PCNA无显著差异,微绒毛结构正常;上述4组胎盘PCNA 显著高于模型组,模型组微绒毛结构消失;模型组HB-EGF不表达,动情期子宫内膜HB-EGF显著高于上述 4组;PCNA与HB-EGF正相关。结论(1):HB-EGF和早孕期滋养细胞的增殖明显相关。(2)中药可能通过 自分泌或旁分泌增加滋养细胞HB-EGF表达,增强滋养细胞的增殖及侵入和对缺氧的耐受。
Purpose To study expression and correlation of HB-EGF(Heparin-binding epidermiz growth factor),PCNA(proliferating cell nuclear antigen) and influence of proliferation on trophoblast with Chinese Herbs for spontaneous abortion. Methods Bromocriptine-induced hypoprolactin rat abortion model was treated by Chinese Herbs, progesterone and PRL respectively, controls were pure model, normal pregnancy and pseudopregnancy rats, each group contained 12 rats. All rats were killed at pregnant day 12 to observe outcome of pregnancy. Expression and correlation of HB-EGF mRNA and PCNA in trophoblast and endometrium was analyzed by RT-PCR and immunohistochemistry. Microvillus was observed by electron microscope. Results There were no significant differences between expression of HB-EGF and PCNA on trophoblast in the model treated by Chinese Herbs, progesterone, PRL and normal pregnancy(P>0.05) . The microvillus was normal in the four groups. Expression of PCNA on trophoblast in these four groups were significantly higher than those of the pure model. Microvillus disappeared in the pure model.There were no expression of HB-EGF on the pure model. Expression of HB-EGF on the oestrum endometrium were significantly higher than those of four groups.There was positive correlation between PCNA and HB-EGF(r=0. 262,P<0.01). Conclusions HB-EGF was correlated to proliferation of trophoblast in early pregnancy. Chinese Herbs may enhance expression of HB-EGF on trophoblast through autocrine and paracrine role,and may enhance proliferation, invasion and tolerance anoxia of trophoblast.
