槲皮素逆转HL-60/ADR多药耐药机制的研究 被引量:4

摘要 目的探讨槲皮素在体外逆转耐药细胞株HL 6 0 /ADR的多药耐药。方法以HL 6 0 /ADR细胞为研究对象 ,用RT PCR法检测耐药基因MRP1的表达及槲皮素对其表达的影响 ;采用共聚焦激光扫描显微镜观察槲皮素作用前后柔红霉素 (DNR)在耐药细胞HL 6 0 /ADR内亚细胞结构分布的改变。结果HL 6 0 /ADR中有MRP1的过量表达 ,且槲皮素能下调该基因的表达 ;槲皮素能恢复DNR在HL 6 0 /ADR细胞中的异常分布 ,回归其作用靶点 ,逆转耐药。结论DNR在耐药细胞中的异常分布与肿瘤细胞耐药基因形成有关 ,槲皮素在体外直接抑制MRP1功能 ,恢复DNR在细胞内的分布。 Objective To investigate the effect of multidrug-resistance reversed (MDR) by quercertn in HL-60/ADR cell line in vitro. Methods We measure MRP1 expression of HL-60/ADR cell line by RT-PCR and study the relationship between quercetin and MRP1 expression. Using confocal scanning laser microscopy(CSLM), we observe the changes of subcellular distribution of DNR in HL-60/ADR before and after treatment with quercetin. Results MRP1 was overexpression in HL-60/ADRcell line and quercetin exerted a dose- dependent down-regulation on the expression of MRP1 gene. Quercetin restored the abnormal distribution of DNR in HL-60/ADR, made DNR return to their targets and reversed multidrug-resistance. Conclusion Altered subcellular distribution of DNR in resistant cell line was related to MDR gene formation. Quercetin could inhibit MRP1 function and could restore the subcellular DNR distribution.
