分泌抗人胎盘型谷胱甘肽S-转移酶单克隆抗体杂交瘤细胞株的建立及鉴定

摘要 通过B淋巴细胞杂交技术,成功地制备了分泌抗GST-π单克隆抗体的杂交瘤细胞株A_1和A_2。对获得的两株杂交瘤细胞株进行了染色体核型分析和DNA含量测定。结果表明,它们确系NS-1小鼠骨髓瘤细胞与小鼠脾细胞的杂交体,抗体类型鉴定结果表明,杂交瘤A_1只分泌IgG_1类免疫球蛋白,而A_2只分泌IgG_(2a)类免疫球蛋白,且两类免疫球蛋白均由k型轻链组成。两株杂交瘤分泌的单克隆抗体仅与GST-π特异反应,与GST的其它类型同功酶及其它可溶性抗原均无交叉反应。 Two hybridoma cell lines, which secreted monoclonal antibodies(McAbs) constantly, were obtained through cell fusion, screening and cloning. The numbers of the chromosomes and the content of the DNA of the two hybridoma cells were determined. The results show that the two hybridoma cells is of the fusion from NS-1 mouse myeloma cells and mouse spleen cells. One of these two hybridomas secretes immunoglobulin G_1 class, whereas the other one secretes immunoglobulin G_(2a), class. All the both McAbs consists of light chain к and specifically recognized the GST from human placenta(class π), showing no cross reactivity against another GST isoenzymes and soluble antigen.
