摘要
目的 探讨端粒长度及端粒酶活性在大肠癌发生发展中作用。方法 采用Southernblot及端粒重复扩增 (TRAP)法检测端粒长度和端粒酶活性水平。结果 大肠癌端粒酶活性明显高于癌旁组织及正常大肠黏膜组织 ;大肠癌组织端粒长度较癌旁组织及正常大肠黏膜明显缩短 ,且随大肠癌Dukes分期的进展进一步缩短。结论 端粒的短缩和端粒酶的激活可能对大肠癌的发生发展起重要作用.
Objective To explore the role of telomere length and telomerase activity in colorectal carcinoma.Methods Telomere length was measured by Southern blot,and telomerase activity detected by TRAP.Results The activity of telomerase in colorectal cancer tissues was significantly higher than that in paracancerous tissues and normal sucoma.The length of telomere in cancer tissue was much shorter than in the adjacent tissues and normal sucoma,and telomere length was decreased gradually along with the development of cancer stage.Conclusion Progressive shortening of telomere and the activation of telomerase probably play a critical role in the pathogenesis of colonic carcinoma.
