摘要
目的 :观察不同恶性级别脑胶质瘤中端粒酶活性及P gp糖蛋白表达的相关性。 方法 :采集手术中切除的不同级别脑新鲜胶质瘤标本 2 6例 ,脑外伤内减压脑组织 8例 ,用TRAP法及免疫荧光染色流式细胞仪法检测不同级别脑胶质瘤端粒酶活性以及P gp糖蛋白表达。结果 :端粒酶活性阳性表达率为 88.5 % ,P gp糖蛋白阳性表达率为 6 5 .4 % ;端粒酶活性阳性病例中 ,P gp糖蛋白阳性表达率 6 9.6 % (16 / 2 3) ,端粒酶活性阴性病例中P gp糖蛋白阳性表达率 33.3% (1/ 3) ,两者差异具有显著性 (P <0 .0 5 )。对照组脑组织端粒酶活性及P gp糖蛋白阳性表达均为阴性。结论 :脑胶质瘤中 ,端粒酶活性和P gp糖蛋白阳性表达具有相关性 ,可将二者结合起来评估脑胶质瘤恶性程度和预测化疗效果。
Objective: To investigate the correlation between the telomerase activity and the expression of p-glycoprotein in gliomas with different malignant degrees.Methods:The activity of telomerase was examined by TRAP method, and the immunohistochemical staining and FLM methods were used to measure P-gp expression in 26 fresh gliomas and 8 normal brain tissues. Results: The telomerase expression was positive in 23(88.5%)glioma specimens,while the positive rate of P-gp was 65.4%. The positive rate of P-gp was 69.6%(16/23) in cases with positive telomerase expression, while 33.3%(1/3) in negative cases, with significant difference between them(P<0.05). Both telomerase activity and p-glycoprotein were negative in normal brain tissue. Conclusions:The level of telomerase activity in gliomas was correlated with the positive rate of p-glycoprotein which represented MDR. High level telomerase activity and high positive rate of p-glycoprotein were detected in malignant gliomas, so they could be taken as the markers of malignant glioma.
