摘要
目的 :为恢复失去卵巢功能妇女体内性激素的作用 ,本实验将大鼠卵巢异体皮下移植 ,观察移植效果。方法 :分别将SD大鼠新鲜卵巢及培养后卵巢异体皮下移植。通过观察阴道脱落细胞、子宫重量的变化 ,移植物的组织形态学及测定血清雌二醇 (E2 )水平了解卵巢移植鼠体内激素分泌状况。结果 :新鲜移植组 ,3/ 10只受体鼠阴道脱落细胞显示有雌激素作用 ,子宫重量及血清E2 显著高于去势组 ,移植物表面无明显血管网形成 ,组织切片可见大量原始卵泡和及少量发育卵泡。培养后移植组 ,6 / 10只受体鼠阴道脱落细胞显示有雌激素作用 ,子宫重量及血清E2 水平显著高于去势组 ,与新鲜移植组无显著差别。移植物表面可见少量微血管分布 ,组织切片可见较多的发育卵泡。结论 :异体皮下移植可降低组织的免疫源性 ,而经体外培养后可进一步降低免疫源性 ,利于移植物存活 ,为临床组织器官同种异体移植提供实验依据。
Objective:In order to recover the hormone function of women who have lose their ovarian function , varian is to be subcutaneous allogenic transplanted ,we observe the effect of this transplantation. Method: Adult female SD rats are used in this study. They are randomly divided into 2 groups:group A-Oophorectomize animals receiving fresh ovarian allognic transplants immediately .Group B-Oophorectomize animals receiving cultivated ovarian allognic transplants .To understand hormone secrection in ovarian transplantation rats and judge their endocrine function by the change of vaginal drop cells and uterine weights, histological morphologic of grafts and the level of the serum E 2.Result:In group A, according to the change of change of vaginal drop cells,3 of 10 animals have the effect of E 2.The uterine weights and the level of the serum E 2 are significantly higher than OophorectomizeAnimals. There are no capillary network in the surface of transplants. There are a lot of primordial follicles and a few developing follicle in tissue slice. In group B, 6 of 10 animals have the effect of E 2.The uterine weights and the level of the serum E 2 are significantly higher than Oophorectomize animals. There are few capillary network in the surface of transplants.There are a lot of developing follicle in the tissue slice. Conclusion:The immunogenicity may decline by the allogenic subcutaneous transplantation ,and those were decline further by culture. These will be benefit for the transplant living and provide the experimental evidence to clinical ovarian transplantation.
