摘要
[目的 ]探讨乙二醇 (EG)、对苯二甲酸 (TPA)联合肾损伤作用。 [方法 ]以SD大鼠EG、TPA经口灌胃亚慢性联合染毒 12周 ,EG单独染毒浓度分别为EGlow:60 0mg/(kg·d)、EGhigh:180 0mg/(kg·d) ;TPA单独染毒浓度分别TPAlow:5 0 0mg/(kg·d) ,TPAhigh:2 0 0 0mg/(kg·d) ,联合染毒则为EG1/ 2low+TPA1/ 2low:3 0 0 +2 5 0mg/(kg·d)、EGlow+TPAlow:60 0 +5 0 0mg/(kg·d)、EG1/ 2high+TPA1/ 2high:90 0 +10 0 0mg/(kg·d) 3个剂量组。对照组给予食用油。收集血、尿进行肝肾功能检查并观察肾组织光镜病理改变和电镜超微结构改变。 [结果 ]TPA和EG染毒组血清尿素氮 (BUN)、肌酐 (SCr)和尿蛋白(UALB)水平显著高于对照组 ,TPA与EG两因素对大鼠血清BUN、SCr和UALB的增高存在联合作用 ,表现为增强作用 (相加或协同 ) ;TPA单独染毒大鼠肾脏病理改变较轻 ,见肾小管蛋白管型浸润 ;EG染毒和联合染毒大鼠肾小管上皮细胞坏死脱落 ,管腔扩张甚至崩解 ,腔内充满草酸钙结晶 ,病变累及到肾小球和肾间质 ,引起肾小球萎缩和肾间质纤维化。 [结论 ]EG、TPA能对肾脏功能产生影响 ,不但影响肾小管功能 ,而且影响肾小球功能 ,在一定剂量下两者对肾脏毒性存在联合作用 。
Objective To investigate its nephrotoxic effects of ethylene glycol(EG),combined with terephthalic acid(TPA) and possible mechanism in animal experiment. SD rats were administrated EG and TPA by gastric gavage,the administrated doses of EG were 600 mg/kg·d, 1 800 mg/kg per day,and the doses of TPA were 500 mg/kg,2 000 mg/kg per day,the mixture of EG and TPA were 300+250 mg/kg,600+500 mg/kg,900+1 000 mg/kg per day,respectively. It has been shown that after 12 week-gavage of EG and TPA,the level of BUN,Scr and UALB was significantly higher in exposed group compared to the control.The EG and TPA had combined toxicity based on the level of BUN,Scr and UALB.At the same time,increased protein casts were observed in TPA-exposed rats,renal tubule expanded,collapsed and necrotic.calcium oxalate congregated which even induced glomerular atrophy and renal interstitial fibrosis in rats exposed to high dose of EG,changes in renal pathological structure were more serious in rats exposed to mixture of EG and TPA. [Conclusion] EG and TPA exposure could induce the renal dysfunction,not only renal tubule damage,but also glomerular damage.And at certain dose,additire or synergistic effect might be observed in kidney.
