摘要
肌苷又称次黄嘌呤核苷,在医疗及食品等方面具有重要用途。目前,我国肌苷发酵水平较低,在工业生产上仅为10g/L 左右,如何提高肌苷生产菌的产苷能力是当前一个重要研究课题。
Taking Bacillus subtilis 7171-9-1 as starting strain,mutants N_(5(?))(Ade^-+Xan^-+SG^r)and N_(168)(Ade^-+His^- +8-AG^r)are obtained after 60 minutes by mutagenic treatmentwith 1% DES.Using N_(5(?)) and N_(168) as parents,a higher inosine productivity of F22 is gainedby protoplast fusion technique.The optimum conditions of protoplast's preparation and re-generation have been determined.Under these conditions,the forming rate and the regenera-tion rate of the protoplasts of N_(58) and N_(168) are 94.7% and 20.9%,91.7% and 24.4%,respec-tively.The fusion rate of protoplast is 5.4×10^(-4).The best conditions of flask-shaking fermen-tation for F22 have also been determined and level of its inosine accumulation is 24.58 g/l.F22has been found to be comparetively stable after 10 generations in flask-shaking,therefore,ithas a higher potential value of application.
