摘要
本实验用HPLC法测定人血浆中的法莫替丁浓度。血浆用硅胶小柱预处理。色谱柱为Spherisorb C_(18)(5μ,4.6×250mm),流动相为甲醇—乙腈—水—磷酸(10V:10V:80V:0.08V)。检测波长为267nm,用外标法峰面积定量(y=0.00805X+3.05,r=0.999,线性范围:12.5~125ng/ml)。回收率下均为80%,日内及日间误差均小于7%。本法灵敏度高,准确性好。适用于生物利用度和临床研究。
A selective and sensitive analytical method was developed for the quantification of famotidine in plasma samples. A straight forward high-recovery sample prepartion procedure was effected by using silica-gel cartirdges to adsorb the hydrophilic drug from plasma.The described sample preparation scheme permitted the quantification of famotidinein plasma by UV detection (267nm) alone. Resolution of famotidine from endogenous interferences was accomplished on a spherisorb C18 column with a methanol-acetonitrile-water-phosphoric acid (10:10:80:0.08)mobile phase.
