皮肤撕脱伤动物模型中整合素CD18的动态表达及其变化规律

摘要 目的:研究整合素CD18在皮肤撕脱伤动物模型中的表达规律,探讨其在组织继发性坏死中的作用。方法:复制皮肤撕脱伤动物模型,分别于0,2,6,12,24h采集组织标本,用PCR反转录扩增的方法检测组织中CD18mRNA,进行半定量测定,了解其随时间的变化规律。结果:①撕脱组中CD18mRNA的含量随撕脱时间的延长而升,12h左右达峰值,此后随时间的延长而略有下降。②撕脱组中CD18mRNA的含量明显高于对照组,统计分析二者差异显著。结论:黏附分子CD18在皮肤撕脱伤早期,表达明显增高,其高表达可能是早期造成撕脱皮瓣继发性坏死的重要原因之一,为临床皮肤撕脱伤的救治和康复提供理论依据。 AIM:To investigate the expressional regularity of integrin CD18 in animal mode ls of skin avulsion and probe into the effect of CD18 on secondary tissue necros is. METHODS: The animal model of skin avulsion was reproduced, and RT PCR was emp loyed to detect CD18 mRNA in tissues at 0, 2, 6, 12, and 24 hours after trauma, and the semi quantitative assay was employed so as to investigate the changing regularity of it along with time. RESULTS: ①The content of CD18 mRNA was gradually increased with the time prol onged and reached its peak value at 12 hours in the avulsion groups, and decreas ed with the time prolonged. ②The content of CD18 mRNA in the avulsion group was obviously more than that of the control group. There was a significant differen ce between them with statistical analysis. CONCLUSION: The expression of CD18 significantly increases in the early period of skin avulsion injury. The high expression of CD18 is possibly one of importa nt reasons for avulsed tissue secondary necrosis. It provides a theoretical basi s for clinical treatment and rehabilitation of skin avulsion.
