摘要
目的探讨日本血吸虫Sj26基因转染对树突状细胞(DC)功能的影响。方法利用脂质体介导的基因转染技术将日本血吸虫Sj26基因转染DC,流式细胞术(FACS)检测Sj26基因转染的DC摄取抗原的能力,混合淋巴细胞反应检测Sj26基因转染的DC对同种异体T淋巴细胞的刺激作用。结果Sj26基因转染后Sj26蛋白在DC中高表达,摄取抗原的能力降低,与pcDNA3转染的DC和未转染的DC相比,Sj26基因转染的DC能明显刺激混合淋巴细胞反应。结论Sj26基因转染DC后能促进DC的活化,增强DC的生物学活性。
Objective To explore the effect of Schistosoma japonicum Sj26 gene transfer on the functions of dendritic cell (DC) in vitro.Methods DCs were transfected with recombinant plasmid pcDNA3-Sj26. The ability of DC to take in antigen was determined by FACS. The mixed lymphocyte reaction was used to detect the effect of Sj26 gene-transferred DC on stimulating allogenetic T lymphocyte. Results Sj26 was expressed at high level in Sj26 gene-transferred DC. Compared with pcDNA3 transferred DC and untreated DC, Sj26 gene-transferred DC had strong potential to stimulate proliferation of allogenetic T cells. Conclusion Sj26 gene transfer can promote DC activation and enhance the biologic activity of DC.[
