不同方式抽取的新西兰白兔骨髓中成纤维细胞集落形成单位数量差异观察 被引量:1

Observations on the Differences of CFU-F Numbers in Bone Marrow Aspirated in Various Ways from New Zealand White Rabbits

摘要 目的 观察不同方式抽取的新西兰白兔骨髓中成纤维细胞集落形成单位(CFU-F)数量差异。方法通过计数在体外培养条件下定量接种骨髓的CFU-F克隆数量观察了新西兰白兔胫骨、股骨、坐骨上三个穿刺点抽取的骨髓中CFU-F在下述情况中的数量差异:1.同一部位分别抽取不同量骨髓(0.2m1、2m1、2ml、1m1):2.同一部位前后两次(间隔一周)抽取同量骨髓(2m1);3.不同性别;4.不同年龄(4-6个月、1-1.5岁、2.5-3岁)。结果1.与首次抽取骨髓(0.2m1)比较,各穿刺点第二次抽取的骨髓(2m1)CFU—F数量虽略有下降,但变化不明显,继续抽取骨髓(分别为2ml、1m1)其CFU—F值均比前一次抽取的骨髓明显下降。通过检测股骨上穿刺点每次抽取前的骨髓像变化证实这种现象是穿刺部位骨髓被外周血稀释的结果:2.胫骨骨髓中CFU-F数量略低于股骨和坐骨骨髓;,3.同一部位后次抽取的骨髓CFU-F数量明显低于前次;4.雄性兔骨髓CFU-F数量略低于雌性兔,但差异无显著性;5.上述三个年龄组骨髓中CFU-F数量无明显差异。结论在研究兔骨髓基质细胞时,若实验要求应用较为纯净的骨髓,抽取骨髓量应在2m1以内为宜或多处取材,避免短期内在同一部位重复取材。 Objective To observe the differences of colony forming unit-fibroblastic(CFU-F) numbers in bone marrow aspirated in various ways from New Zealand white rabbits. Methods Marrow were obtained by aspiration from the three puncture points (pp) at tibia, femur, is chium of New Zealand white rabbits respectively and the CFU-F numbers of the marrow specimens were compared by enumeration of the CFU-F clones in culture under the following conditions: thirty two rabbits were assigned to four groups, that is, ① Different amounts of marrow (0.2ml, 2ml, 2ml, 1ml) aspirated from each pp of the three bones. ② Equal amounts of marrow (2ml) aspirated two times from the same pp of the three bones at an interval of one week. ③ Equal amounts of marrow (2ml) from the male and the female rabbits. ④ Equal amounts of marrow (2ml) from the rabbits of different ages (4-6 months, 1-1.5years, 2.5-3years). Results ① In group 1, although the CFU-F number of marrow aspirated at the second time (2ml) was slightly lower than that at the first time (0.2ml), there was not significant difference between them, whereas the CFU-F numbers of marrow aspirated at the third and the fourth times (2ml, 1ml) were both evidently lower than those at the second and the third time respectively. This phenomenon was attributed to the marrow specimens diluted by peripheral blood. ② In group 1, the CFU-F number of marrow from tibia was slightly lower than those from femur or ischium. ③ In group 2, the CFU-F number of the marrow from the three pp aspirated at the second time were significantly lower than that at the first time. ④ CFU-F number of marrow from the female was slightly higher than that from the male. ⑤ In group 4, there were not significantly differences among the CFU-F numbers of marrow from the rabbits of three age groups. Conclusions When studing marrow stromal cells of rabbit for obtaining relatively pure marrow if experimental requirement, marrow aspirated should be limited to less than 2ml or aspiration of it at two or multiple
