摘要
该文研究了幽门螺杆菌(Helicobacterpylori,简称HP)疫苗对母鸡诱导产生特异性抗体(Immunoglobulinofyolk,简称IgY)以及IgY的制备和检定方法.采用融冻法和超声波法对HP破碎,破碎后的细胞用福氏完全佐剂和福氏不完全佐剂制成疫苗对生蛋母鸡进行免疫,通过ELISA间接法检测鸡蛋的免疫应答,IgY用盐析、超滤和葡聚糖凝胶(SephadexG 200)层析等方法纯化,未纯化和纯化的IgY进行SDS PAGE电泳分析.IgY与HP的凝集反应均为阳性,表明IgY为HP的特异性抗体,用ELISA测定效价达1.6×103,比未免疫的蛋黄IgY高约160倍,根据SDS PAGE图谱分析抗HP的特异性IgY分子量在180kD左右.
Preparation and identification of egg yolk immunoglobulin against Helicobacter pylori (HP) are studied. HP by frozen-thaw and ultrasound was made to bacterin with the Freund's incomplete adjuvant and Freund's complete adjuvant. Chickens were procedurally immunized with the bacterin. A special immunoglobulin against HP was prepared from immunized egg yolk with saltingout, column chromatgraphy and ultra-filtration; Purified immunoglobulins (IgY) was identified for SDS-PAGE with the Helicobacter pylori. High titer egg yolk immunoglobulins were produced by means of procedural immunization with HP. The ELISA titer was measured at 1.6×10~3 and the figure of SDS-PAGE had revealed that the molecular weight of the purified immunoglobulin against HP is about 180 kD. Agglutination test has shown that prepared IgY can effectively react with the HP.
