彩超照射后胎鼠耳蜗毛细胞的定量及形态学观察 被引量:1

A Quantitative and Morphological Study on Cochlear Hair Cell Underwent Diagnostic Ultrasound Exposure in Fetal Guinea Pigs

摘要 目的 通过琥珀酸脱氢酶 (SDH)组化染色和图象分析和耳蜗基底膜扫描电镜观察来评估彩色多普勒超声检查 (彩超 )对胎鼠耳蜗毛细胞的影响。方法 彩超分别照射受孕前期 (前 15d)及受孕后期 (4 0~ 5 5d)豚鼠子宫 ,5min/次 ,1次 /d。通过对毛细胞形态学观察和毛细胞记数定量分析彩超对胎鼠听觉发育的影响。结果 ⑴实验组与对照组相比 ,毛细胞计数有显著差异 ,其中实验组以第 2回 ,第 3回和第 4回损害严重 ,与对照组相比差异非常明显。⑵受孕前期照射组与受孕后期照射组相比较 ,毛细胞计数无统计学显著差异。⑶扫描电镜下毛细胞损害明显 <SDH染色铺片毛细胞的损害数目。结论 ⑴医用超声在一定照射时间下影响胎鼠毛细胞发育。毛细胞损害以耳蜗底部区域为主。⑵超声对受孕前期胎鼠的毛细胞损害程度同于受孕后期。⑶超声波使琥珀酸脱氢酶 (SDH)活性降低的程度高于细胞表面形态学改变。 Objective To investigate the effect of diagnostic ultrasound on cochlear hair cells in fetal guinea pigs. Methods Diagnostic ultrasound was used to irradiate the uteri of early pregnant guinea pigs(the first 15 days of gestion) and late pregnant guinea pigs(the 40th to 45th days of gestion) for 5 minutes daily. The number of cochlear hair cells in fetal guinea pigs was calculated using succinic dehydrogenase (SDH) staining, and computer image analysis system. The morphological change of cochlear hair cells was observed with scanning electron microscope(SEM). Results Compared with non-exposed control group, the number of total hair cells obviously decreased in the two exposed groups (P<0.05), especially after the second, third and fourth times of irradiation(P<0.01). There was not significant difference in the number of total cochlear hair cells between the early and late exposed groups. In the exposed groups, the number of damaged cochlear hair cells observed with scanning electron microscope was less than that observed by SDH staining. Conclusion Diagnostic ultrasound for certain times could damage developmental cochlear hair cells in fetal guinea pigs, especially ones at the cochlear bottom. The impairment of diagnostic ultrasound on the cochear hair cells of early stage pregnancy was the same as that of late stage pregnancy. The degree of SDH level decrease caused by diagnostic ultrasound irradiation was higher than that of cochlear hair cells surface change.
