心肌超声造影技术评价骨髓细胞移植于缺血心肌的促微血管新生效应 被引量:3

Myocardial contrast echocardiographic assessment of neovascularization of ischemic myocardium by bone marrow stromal cell implantation in rabbit

摘要 目的探讨骨髓间质干细胞移植到缺血心肌后局部新生的微血管在活体心脏的灌注状况。方法对正常、心肌缺血梗死和骨髓细胞移植兔进行心肌超声造影(MCE)检查,评价缺血梗死区造影剂填充程度及强度。免疫荧光技术检测移植区微血管新生状况。结果正常时兔心肌内造影剂均匀分布,急性心肌梗死后左室前间壁造影剂充盈缺损,而移植4周后该处心肌内可见少量造影剂回声。定量分析显示移植组造影剂峰值强度明显高于心肌缺血梗死组[(44±20)dB对(17±5)dB,P<0.01]。移植区可检测到Brdu标记阳性的移植细胞抗Ⅷ因子染色阳性,毛细血管密度较缺血梗死组显著增高(P<0.05)。结论骨髓细胞移植可导致缺血心肌局部功能性微血管新生,心肌组织灌注状况改善。 Objective To investigate focal myocardium microvascular perfusion in vivo after bone marrow stromal cells (MSCs) transplantation into ischemic region by myocardial contrast echocardiography (MCE) in rabbits′ heart. Methods MCE was executed on normal, myocardial infraction(MI) and MSCs implantation rabbits, and the perfusion degree and peak intensity(PI) of microbubbles were detected. Anti-Ⅷ factor staining and vascular density were evaluated immunohistochemically to assess the angiogenesis effect. Results Microbubbles were located homogenously in normal myocardium, but obvious contrast defection could be seen in MI rabbits. In MSCs treated rabbits, spotted contrast echo could be observed and PI was significantly increased when comparing with MI group [(44±20)dB vs (17±5)dB,P<~0.01 ]. Bromodeoxyuridine (Brdu) labeled cells were found in ischemic region, companied with positive anti-Ⅷ factor staining and significantly enhanced vascular density (P<~0.05 ). Conclusions MSCs implantation induces angiogenesis in rabbit focal myocardium and improves blood perfusion in vivo.
