摘要 目的初步探讨瘦素(Leptin)对成骨细胞骨保护蛋白(OPG)mRNA表达的影响。方法用SYBR Green Ⅰ染料实时监测定量RT-PCR的方法测定骨保护蛋白(OPG)mRNA表达。结果 瘦素(Leptin)使成骨细胞骨保护蛋白OPG mRNA表达呈剂量和时间依赖性降低。结论 瘦素(Leptin)作为一种内分泌因子,可直接与成骨细胞上leptin受体特异结合,通过受体后信号转导下调节成骨细胞OPG mRNA表达,使OPG mRNA表达降低,和其它因子共同参与骨再建调节。 Objective To study the effect of leptin on the expessione of osteoprotegerin (OPG) mRNA in osteoblast-like cells. Methods The expression of OPG mRNA in osteoblast-like cells was determined by real-time quantitative RT-PCR based on Light Cycler technology. Results Leptin down-regulated the mRNA levels of OPG (P<0.05) in a dose-and time-dependent manner. Conclusions Leptin acts directly on osteoblasts to regulate bone remodelling by decreasing OPG mRNA.
